DLL3（Delta-like ligand 3）是Notch信号通路的非典型配体，属单次跨膜蛋白，其胞外域含6个EGF样重复序列，但缺乏与Notch受体（如NOTCH1）直接激活所需的DSL（Delta/Serrate/LAG-2）结构域，通过反式或顺式结合抑制Notch信号传导，导致下游靶基因（如HES1）表达下调，促进神经内分泌分化及肿瘤细胞去分化。功能上，DLL3在正常发育中调控神经前体细胞极性，但在恶性肿瘤如小细胞肺癌（SCLC）、大细胞神经内分泌癌（LCNEC）及胶质瘤中异常高表达（>80% SCLC患者），且与肿瘤干细胞特性及化疗耐药相关。 本品为靶向DLL3重组抗体(Rovalpituzumab)。 |
